This <br /> <br /> experiment aims to test the antibacterial activity of ethanol extract from leaves and stem bark of <br /> <br /> Nyamplung against the Propionibacterium acnes and Staphylococcus epidermidis. Extraction was <br /> <br /> carried out by the reflux method using ethanol as a solvent. The selected extract was fractionated using <br /> <br /> liquid-liquid extraction with n-hexane and ethyl acetate as solvents. Diffusion method and <br /> <br /> microdilution were used to test antibacterial activity. From the preliminary test, at a concentration of <br /> <br /> 10% ethanol extract of the bark had a largest inhibit zone against Staphylococcus epidermidis with <br /> <br /> diameter inhibition 11.68 ± 0.81 mm. Microdilution test results of ethanol extract, water fraction, and <br /> <br /> ethyl acetate fraction of stem bark had MIC (Minimum Inhibition Concentration) values with the range <br /> <br /> from 1024 - 2048 μg / mL and MBC (Minimum Bactericidal Concentration) values with the range from <br /> <br /> 3125 - 6250 μg / mL against Staphylococcus epidermidis, while the n-hexane fraction showed no <br /> <br /> antibacterial activity. The stem bark extract and fractions were tested in bioautography. Bioautography <br /> <br /> showed that the compounds contained in the water fraction with the Rf value of 0.2 on the silica gel <br /> <br /> GF254 TLC plate with the mobile phase of chloroform-ethyl acetate-formic acid (1: 3: 1) gave inhibitory <br /> <br /> areas against S. epidermidis.
