Description
Summary:
Beauveria bassiana produces some insecticidal metabolite. It can be used as a biological alternative pesticide agent beside the chemical one. The aim of this study is to determine the larvicidal effect of an ethyl acetate and a hexane extract from B. bassiana filtrate. In this work, firstly, the incubation time to harvest a spore from potato dextrose agar (PDA) is defined according its number and viability. Secondly, the time to get a filtrate or fermentation product from potato dextrose broth (PDB) that give the highest mortality against second instar Aedes aegyti larvae is determined. Finally, liquid-liquid extraction (1:1) from the filtrate is done. The solvent phase is separated and evaporated to get precipitate of extract from each solvent. To determine the value of LC50 (lethal concentration that kills 50% of the exposed larvae), the concentration extraction product is adjusted with dimethyl sulfoxide (DMSO). The bioassay is done against second instar Ae. aegyti larvae. The incubation time to harvest the spore from PDA medium is ninth day old culture with the spore number and the viability is 2.54 x 107 spore/mL and 93.46%, respectively. The filtrate of sixth day old fermentation in PDB medium produce filtrate that give 100% mortality against second instar Ae. aegyti larvae, the highest mortality. LC50 value for 3 days of ethyl acetate and hexane extract is 117.28 dan 287.09 ppm, respectively. This result shows that the ethyl acetate and hexane extract of B. bassiana filtrate have biopesticide potential.
