cause of death in the world. Pomegranate (Punica granatum L.) has been used for a <br />
<br />
long time to treat various diseases, including inflammation, which is expected to be an <br />
<br />
alternative therapy. Screening the pharmacological activities of active ingredients and <br />
<br />
knowing the mechanism of its interaction with the target protein is an expensive <br />
<br />
process and requires a long time if only done using experimental animals. So this study <br />
<br />
aims to predict the interaction of compounds contained in pomegranate peels (Punica <br />
<br />
granatum L) against cyclooxygenase in in silico, test the toxicity of water fraction, and <br />
<br />
also test the anti-inflammatory of ethanol extract and water fraction in zebrafish <br />
<br />
embryos. Interactions are predicted by docking to cyclooxygenase-1 (PDB ID: 5WBE) <br />
<br />
and cyclooxygenase-2 (PDB ID: 5IKR). Toxicity test was carried out based on OECD <br />
<br />
number 236, while the anti-inflammatory test of zebrafish embryos was carried out by <br />
<br />
staining using neutral red at five different concentrations induced by carrageenan. The <br />
<br />
best compounds by docking are punicic acid and kaempferol, have a better interaction <br />
<br />
than native ligands with binding energy -7.7619, -7.3755 at 5WBE and -8.3216, - <br />
<br />
7.1262 at 5IKR, respectively. Lethal Concentration 50 (LC50) of water fraction is <br />
<br />
236.57 ± 6.1 µg / mL, categorized as safe according to the environmental safety <br />
<br />
category of the OECD. Whereas the results of anti-inflammatory testing are observed <br />
<br />
based on fluoroscence intensity, survival rate, and heart beat rate, with the best <br />
<br />
concentration result on ethanol extract is 23.6 µg/mL and water fraction is 21.6 µg/mL. <br />
<br />
So it can be concluded that ethanol extract and water fraction of pomegranate peels <br />
<br />
have the potential as candidates for anti-inflammatory drugs. <br />
