GLYCOSIDES DITERPENE COMPOUNDS (ENT-KAURENE GLYCOSIDES) AS SECONDARY METABOLITES FROM STEVIA REBAUDIANA BERTONI LEAVES

Stevia rebaudiana Bertoni is species of the genus Stevia. This plant is known as a natural sweetener and known as sweetleaf, sugarleaf or sweet herb. This designation is given because these species have the high sweetening potency that is about 50-500 times compared with sucrose sugar...

This plant is known as a natural sweetener and known as sweetleaf, sugarleaf or sweet herb. This designation is given because these species have the high sweetening potency that is about 50-500 times compared with sucrose sugar that has been widely used. This plant is also used as a medicine as a cure for diabetes mellitus, hypertension drug, and can slow the formation of plaque and dental caries. The main components of this species is a class of compounds diterpene-glycosides (ent-kaurene glycosides) that is stevioside, rebaudioside, and dulcoside. On the leaves, there are some classes of compounds reported as labdane, flavonoid, sterols, and triterpenoids. In this research, the isolation of compounds from the leaves of Stevia rebaudiana Bertoni is done using maceration method with methanol solvent. Then, the liquid-liquid extraction with the solvent n-hexane, ethyl acetate, water, and n-butanol. Butanol fraction was separated and purified by two methods, recrystallization method and chromatography methods (vacuum and press) using n-hexane, ethyl acetate, and methanol as solvent with increasing polarity. The results were analyzed with IR spectroscopy, 1H NMR spectroscopy and 13C NMR spectroscopy. Isolation of pure compounds results was obtained a white solid (5 g) is suggested as stevioside with yield 5% and brownish red paste (7mg), which is still a group of diterpene-glycosides compounds with yield 0,21% of the weight of dry leaves. text
