Summary: | This research was conducted with the aim of increasing the extraction of lutein and zeaxanthin compounds from the leaves of sweet potato plants of the Cilembu variety with by meant of pre-treatments of leaves using the Aspergillus niger mushrooms. In this research there are two variations, there are shade drying time of sweet potato leaves, 3 days, 6 days and 9 days, and fermentation time of 4 days, 7 days, and 10 days. The biological agent used in the pre-treatment of fermentation was the Aspergillus niger mushrooms. The results of the drying treatment of the shade drying of leaves showed that drying for 9 days resulted in the highest extraction of lutein and zeaxanthin compounds, respectively 0.934 g / g dry substrate and 0.029 g / g dry substrate. The results of variations in leaf pre-treatment fermentation showed an increase in the acquisition of lutein and zeaxanthin compounds up to 18.57% for lutein and 3.39% for zeaxanthin compared to the acquisition of lutein and zeaxanthin compounds from Cilembu varieties sweet potato leaves without initial treatment of fermentation using A. niger mushrooms. The initial treatment of cellulose degradation with Aspergillus niger fungi with 7 days fermentation resulted in the highest yield of lutein and zeaxanthin compounds, namely 5,030 g / g dry substrate and 0.856 g / g dry substrate, respectively. From the results of fractionation showed a decrease in the components of hemicellulose and cellulose in sweet potato leaves of Cilembu variety due to the hydrolysis activity of cellulase enzyme produced by Aspergillus niger. The pre-treatment fermentation period of 7 days with a low moisture content of leaf has the potential to increase the extracted of lutein and zeaxanthin compounds from sweet potato leaves of the Cilembu variety. |
