alternatives. There are many natural sources that have the potential to produce secondary
metabolites that are bioactive in nature, one of which is endophytic microbes.This study aims to
isolate endophytic fungi that are associated with seagrass originating from Pulau Kaliage Kecil
Kepulauan Seribu and investigate their antimicrobial activity against Escherichia coli ATCC 8739 and
Staphylococcus aureus ATCC 6538 accompanied by determinating MIC and MBC values. This study
consisted of taking seagrass samples then continued with the isolation of endophytic fungi and
fermented. The results of this research were obtained 15 fungal isolates that were coded as FKAK C1
to FKAK C15. Then the mycelium was separated from the culture medium by centrifugation and then
extracted by maceration method while the culture media was extracted by liquid-liquid extraction
method using ethyl acetate solvents. Each extract obtained from mycelium and culture media was
tested for antimicrobial activity by disc diffusion method and then determined the minimum
inhibitory concentration (MIC) value with microdilution method and minimum bactericidal
concentration (MBC) with growth methods on agar media. The greatest antibacterial activity was
showed by mycelium culture of FKAK C5 extract (64 µg/ mL), mycelium culture and culture media of
FKAK C6 extract (64 µg/mL) and mycelium culture of FKAK C11 extract (64 µg/ mL) against
Staphylococcus aureus and extract of culture media FKAK C11 (32 µg/mL) against Escherichia coli. The
greatest antifungal activity was also showed by FKAK C11 mycelium culture of 64 ?g /mL extract. The
lowest MBC value was 64 µg /mL obtained from FKAK C11 mycelium culture extract against
Staphylococcus aureus and extract of culture media FKAK C11 (128 µg /mL) against Escherichia coli.
While the lowest MFC value was 64 ?g /mL for Candida albicans obtained from FKAK C11 mycelium
culture extract.
