Summary: | Enrofloxacin is fluoroquinolone antimicrobial agent which is widely used in poultry. Uncontrolled
application in poultry can accelerate the anti microbial resistency. It is saved in muscle tissue,
skin and fat, liver and kidney. Biological sample has complex matrix. It need both complex and
complicated preparation to produce fast and accurate result. Nowadays, the method of separation
using Molecular Imprinted Polymer (MIP) is developed to solve the analytical problem caused
complex matrix. MIP is a polymer that made using templating method the molecule that has affinity
with template so it can elevate the selectivity and sensitivity. In this study, MIP was made using
enrofloxacin as template molecule, itaconic acid as monomer, etilenglikol dimetakrilat (EGDMA)
as crosslinker, azobisisobutironitril (AIBN) as initiator, chloroform: methanol (4:1) as porogen.
Synthesis MIP did through exaggerate polimeration using molecule ratio of
template:monomer:crosslinker 1:6:20. It produced 1,84 as an IF. MIP that produced was selective
for enrofloxacine. Enrofloxacine assay using HPLC (UV-Vis detector), C18 coloumn and 20 mM
phosporic acid pH 3,0: acetonitrile (83:17) as mobile phase. It produced R2 0,9990 as linearity
test result, recovery between 98,75%-101,84% as the result of accuracy test and RSD 1,83% as
result of precision test. LOD of this method was 1,23 µg/ml and LOQ 4,09 µg/ml. Molecularly
Imprinted Solid Phase Extraction (MISPE) method that appicated on chicken muscle that added
by enrofloxacine produced 8,83±15,19 mg/kg and did not detected on eggs.
