RPO processing is very important to increase selling value and increase the efficacy in its use. To increase its activity, RPO will be formulated as nanoparticle. This study aimed to determine the optimal formulation for RPO nanoemulsion and to test the in vitro antioxidant and inhibition of albumin denaturation activity. RPO nanoemulsion prepared by sonication method. Antioxidant activity was tested by the DPPH (2,2-diphenyl-1-pikrilhidrazil) assay and the result expressed as vitamin E equivalent antioxidant capacity. The test results produced optimal formulations with RPO (5%), Tween 80 (9%), Cremophor RH40 (3%), glycerol (4%), and distilled water (79%). The optimal formula gave globule size at 82.3 nm with PI at 0.385 and stable during the testing period of 28 days. There was an increase in antioxidant activity in RPO nanoemulsion. RPO gave antioxidant activity of 2.35 ± 0.28 ppm vitamin E/mg sample and RPO nanoemulsion gave antioxidant activity of 8.45 ± 0.48 ppm vitamin E/mg sample with the vitamin E as standard gave IC50 of 7.68 µg/mL. RPO nanoemulsion gave an albumin denaturation inhibition activity of 16.43 ± 1.93% for nanoemulsion with 5% RPO concentration and 24.85 ± 1.74% for nanoemulsion with 7.5% RPO concentration. text
