Summary:Arbutin, niacinamide and 3-O ethyl ascorbic acid are compounds that have a whitening effect on the skin and are widely used in whitening creams. Commercial product of whitening cream are generally added preservatives to prevent the growth of microorganisms. Preservatives commonly used are paraben preservatives including methyl paraben, ethyl paraben, propyl paraben, butyl paraben. In monitoring and guaranteeing consumer safety, a specific and selective analytical method is needed to determine the levels of arbutin, niacinamide and 3-O ethyl ascorbic acid without interference from other analytes such as preservatives. Therefore a High Performance Liquid Chromatography (HPLC) method which can analyze these three compounds simultaneously is needed to be developed. In this study, the optimum conditions were obtained by using the Inertsil ODS-3 RP-C18 column, the mobile phase with gradient system and UV detector at a wavelength of 220 nm. The results of method validation showed good linearity with a correlation coefficient (r 2 )> 0.999, VX0 <2%, the accuracy test results also met the requirements with percent recovery of each analytes were 96.93-99.55%, 98.60-99.72%, 97.8-101.6% for arbutin, niacinamide and 3-O ethyl ascorbic acid respectively. Intra-day precision and inter-day precision test result showed the percentage of relative standard deviation (% SD) less than 2% with a HorRat value also less than 2. Limit of detection of this method was 7.07 ppm for arbutin, 19.19 ppm for niacinamide, 7.04 ppm for 3-O ethyl ascorbic acid and limit of quantization was 21.43 ppm for arbutin, 58.15 ppm for niacinamide and 21.33 ppm for 3-O ethyl ascorbate acid. The results showed that the developed method was selective, linear, accurate, precise and can be used to analyze the levels of arbutin, niacinamide and 3-O ethyl ascorbic acid with the presence of paraben preservative in whitening creams simultaneously.
