Local plant of Southeast Sulawesi that is empirically used by the local community in their daily diets but has minimal evidence-based makes Etlingera caloprhys or locally known as olae is one of the local plants that need to be explored, especially as antiobesity. This study aims to determine the activity of ethanol extract of olae as an in vitro antiinflammatory and anti-obesity in vitro and in vivo using zebrafish (Danio rerio) model. The zebrafish were induced using a high-fat diet and the treatment was carried out for 21 days by supplemented olae extract 10% (b/b) and 20% (b/b) into the diet. The antiobesity activity was determined by parameters of body weight, body mass index (BMI), triglyceride levels and blood glucose levels. In vitro test results showed that ethanol extract of olae with a concentration of 600 µg/ml did not show differ significantly (p>0,05) with diclofenac sodium 100 µg/ml in stabilizing membranes from lysis and ethanol extract of olae with varying concentrations have potential inhibiting lipase enzyme activity (dose dependent). The results of the anti-obesity in vivo showed that dietary supplementation of ethanol extract of olae was significantly reduced BMI (p<0,01), body weight (p<0,01), and triglyceride levels (p<0,01) compared with control. Based on the results of this study, flavonoid and phenol compounds of ethanol extract of olae can reduce body weight and triglyceride levels by inhibition of pancreatic lipase and can prevent metabolic syndrome-induced by obesity by prevention of inflammation with a membrane stabilizing mechanism. text
