There are various test methods to quantitatively determine the amount of vitamin B12 in a sample. One of the methods that may be used is microbiological method that utilizes the growth parameter of microorganism as measurement of vitamin B12 activity in a sample. Microorganism that is used is chemo-organotrophic microorganism, which is a microorganism that requires extrinsic nutrition for their growth. Microbiological method that is written in Farmakope Indonesia 5 th edition uses Lactobacillus leichmanii as test microorganism, while this study uses Lactobacillus acidophilus as test microorganism. Lactobacillus acidophilus is one of microorganisms that has chemo-organotrophic properties, so that it may be used as alternatives for determining the activity of vitamin B12. This study aimed to validate the microbiological method of determining cyanocobalamin activity using Lactobacillus acidophilus ATCC 4356. The results of this study showed standard curve linearity with a correlation coefficient of 0,9666, percent recovery ranging from 9.14% to 197.95%, and standard deviation ranging from 0.167% to 63.57%. These results indicated that the validation parameter values obtained did not fulfill the acceptability requirements of a good analytical method validation, thus required further reviews. text
