Herbal medicines for gout which widely sold in the market are types of herbs that have been proven empirically. The purpose of this study was to analyze the methods of measuring uric acid in humans and mice using commercial reagent kits for antihyperuricemia effects and to determine their correction factors. Currently in conducting pre-clinical trials to determine the antihyperuricemia effect, a test using reagent kits is done to measure blood uric acid levels in humans. So far there is no uric acid reagent kit available for rats. This study began with the determination of the peak wavelength of uric acid, and the results showed the peak was at 290 nm with an absorbance of 0.8666. Furthermore, the measurement of absorbance of human and rat serum added with pure uric acid using each of the three samples of human serum and rat in two measurements. This research was conducted in accordance with the standard addition method to see the difference between the serum concentrations of human uric acid and rats. Based on calculations using the standard addition method, serum concentrations of uric acid in humans and mice yielded 4.67 mg / dL and 1.3 mg / dL. From this study it can be concluded that in research on drugs or extracts that use mice, it is necessary to consider the uric acid levels of rats lower than humans because mice have uricase while humans do not have uricase. text
