Many evidences showed that natural antioxidant may be useful in preventing the deleterious consequences from oxidative stress. Based on several researches, the peels of banana (Musa sp.) have been proved as having antioxidant activity. The aims of this research were to determine antioxidant activities of various extracts of tanduk banana peels, total phenolic, flavonoid, and carotenoid content of each extract and analyze their correlation with DPPH-radical scavenging activity; also to isolate antioxidant compound from tanduk banana peels. Tanduk banana peels was extracted using reflux method. Antioxidant activity was determined by 2,2-diphenyl-1- picrylhydrazyl (DPPH) method; phenolic, flavonoid and carotenoid content were evaluated by uv-vis spectrophotometry method; while correlation between total phenolic, flavonoid, carotenoid content and its ??ð??ð¸ ?? ®?ð?ð?ð±??±?± ? ???±¸ ?????± ??????????±?í?¸???????? ®í ±??? ®?? ????ð???±¸ ?thin layer chromatography (TLC). Chosen extract (n-hexane extract) was fractionated by vacuum liquid chromatography (VLC). Purity test of isolate was done using thin layer chromatography (TLC). Isolate was characterized using specific spray reagent, and some pure compounds as comparison. Ethanol extract of tanduk banana peel (density of its 1% extract was 0.84 g/mL) presented the lowest IC50 of DPPH scavenging activity (0.070 µg/mL) and the highest total phenolic content (3.580 g GAE/100 g). The highest total flavonoid content was given by ethyl acetate extract (2.490 g QE/100 g), while nhexane extract gave the highest total carotenoid content (1.288 g BE/100 g). The total phenolic content of tanduk banana peels extracts had negative and significant correlation with their IC50 of DPPH scavenging activities. Phenolic compounds were the main contributor on antioxidant activity of tanduk banana peels. Antioxidant compound which was isolated from n-hexane peels extract of tanduk banana was steroid compound, with IC50 DPPH scavenging activity 4.467 µg/mL. text |
