The current extraction methods for measuring squalene concentration have many limitations, such as require abundant of organic solvents as well as take several times. In addition, the methods are not spesific since other hydrocarbon and sterol (cholesterol) were still mixed with squalene. The purpose of this study is to evaluate quality of shark liver oil capsule on market and improve squalene determination method using Fourier Transform Infrared Spectroscopy (FTIR Spectroscopy). As many as 6 samples were tested for the quality in accordance to SNI through organoleptic test (<7), moisture content (max 0.3%), free fatty acid content (max 1.5%) and squalene concentration (min 70%). The quantitative analysis method were developed using FTIR spectroscopy in wave number 1668.12 cm -1 . The results showed that the level of squalene in all samples do not meet the standard requirement, however all samples meet the free fatty acid content requirements. One sample does not fit the standard criteria in the term of organoleptic test and moisture content. In the other hand, the development of analysis method using FTIR spectroscopy has produced absorbance in proportion to the levels of squalene with correlation coefficient (r) > 0.99, despite of data which does not yet meet the requirement toward validity. This finding suggests to conduct the next research in order to determine squalene concentration using other method such as Raman spectroscopy. text |
