CD can be used to improve the solubility of hidrofobic compound. CD is produced by enzymatic
reaction of the conversion of starch by cyclodextrin glycosyltransferase (CGTase, EC. 2.4.1.19).
í±?± ¬?± ?í?±± ???±??» ?????????íð®í ¬?± ?-???????????¬?¸ ??í± ?í¬??¬®±??ð®¬?ð?¸????????¬?????± ?-
????????¸?± ??ð??????®?????» ???¸?®?ð??¬?¸ ?í± ??ð?¬ð?ð???» ?-CD cavity with many hydrophobic
®??????¸??í± ¬ð??» ?íð????¸?ð???®???¬?± ?í± ?¬?ð???®í¬?¬®?±???» ?-CD which are produce
in 1 litre laboratory scale by the enzymatic reaction using recombinant CGTase (rCGTase) of Bacillus
sp. A2-5a. An amount of 20 UA crude extract and purified rCGTase enzyme was added into 1% sago
??¬?®í ?????¬?± ¬??: ????????????»????í????ð?¬ ??¬?ð® ??¬?±???ð»ð®¬?ð???» ?-CD was including
the isolation by crystallization and purification by adding cyclohexane as complexing agent.
>?ã¬???±??ð® ±?¬??¬?ð???¸±ã?±± ?» ???ð????ð±?¸?¬?¸ ?í± ?¬?ð??» ??????????????±?± determined by UVvis spectrophotometry, high performance liquid chromatography (HPLC), X-ray diffraction (XRD),
electrothermal, and a polarized microscopy. ?-CD produced by rCGTase crude extract showed yield
?» ????????????ð?í ???ð???» ???????????±¬??íð?± ?-CD produced by pure rCGTase gave
yield of 9.08 ± 2.19 % and purity of 95.57 ± 1.35. The results of HPLC analysis showed retension
time of 10-11 ?ð???±???-CD from crude and purified rCGTase which are the same as showed by
standard ?-CD. The results of PXRD analysis gave specific peaks ¬???= 12.62???????and ??????
?íð®í ð??¬?± ¬?¸ð»»?¬®??ã?¬??-CD standard. Melting point analysis using electrothermal for ?-CD
produced by crude extract, by purified rCGTase and for standard ?-CD were 289.1-289.6 °C, 288.9-
289.6 °C and 289.4-289.9 °C, respectively. Polarized microsope images showed that the crystal of
?-CD from this experiment were flat thin shaped which are similar to standard ?-CD.
