The aims of this research are to perform in vitro test of inhibitory activity from lengkuas extract and make lozenge to treat oral Candida albicans infection. The rhizome of lengkuas was extracted with ethanol 96% by maseration process. The extract was concentrated with rotary evaporator. Phytochemistry screening and and quality control of crude drug were observed. Concentrated extract was tested in vitro to determine its minimum inhibitory concentration (MIC) by microdilution method. The concentrated extract was dried and transferred into powder with the addition of avicel PH 101 as an adsorbent. Extract powder then formulated into lozenge by direct compression method. Three lozenge formulas were prepared with different amount of binder. Compressed mass was evaluated and then they were compressed as lozenges. The lozenges were evaluated their organoleptic, friability, hardness, mass uniformity, size uniformity, and disintegration time. Lengkuas rhizome extract contained compounds of flavonoid group, phenol group, and steroid/triterpenoid group. Water contain of extract was 23,55% ethanol soluble matter of 10,97%, water soluble matter of 10,92%, total ash contain of 7,37%, water soluble ash 5,68%, acid insoluble ash 3,78% and loss of drying of 14,62%. Avicel PH 101 was used as an adsorbent because it can provide the powdered extract a good flowability, so direct compression method was applicable. The MIC values of concentrated extract was 400 µg/mL. The best formulation for lengkuas lozenge were 100 mg extract, 500 mg avicel PH 101, 420 mg mannitol, 420 mg sucrose, 4% PVP, 1% magnesium stearate, and 2% talcum because of long residence time inside mouth cavity. text |
