Early aging can be caused by the exposure of UV-light, hormone, and skin nutrition deficiency. Poly-gammaglutamic acid is a substance which is used as a moisturizer which can moisturize and enhance skin elasticity, and reduce skin wrinkles. ?-PGA is an anionic, water-soluble, biodegradable, edible, non-toxic, and non-imunogenic biopolymer. In this research, ?-PGA is fermented by Bacillus subtilis ATCC 6633. The ?-PGA was extracted by using methanol precipitation method and then lyophilized. ?-PGA extract was characterized and used in gel formulation. The evalution of gel dosage form includes gel stability, anti-wrinkle activity, and dermal irritation test. ?-PGA extract occurs as a white odorless fine powder. The fermented ?-PGA extract is water soluble and forms clear solution. The rendemen of ?-PGA extract is 12,497 ± 0,373 g/L. The water content of ?-PGA extract is 4,76±0,022%. The pH of 10% solution is 6,23±0,02, and the pH of 5% solution is 6,67±0,03. The total of carbohydrate contaminant is 2,412±0,102% and the total of ?-amino acid contaminant is 7,618±0,498%. The gel should be clear, has a good viscosity and has pH between 4 and 6,5. The stable gel formulation is obtained with composition of ?????-PGA extract, 0,7% Carbomer, 0,65% triethanolamine, 0,18% methylparaben, 0,02% propylparaben, 5% propylene glycol, and 92,95% aquadest. The final gel formulation is non-irritant to the skin and could reduce roughness index when evaluated by using video dermatoscopy device. text
