Summary: | Many researches have shown that isolated metabolites from marine fungi have antibacterial and
anticancer activity. This research’s purposes were to determine antibacterial activity of isolated
fungi from Pejarakan Mangrove Forest against Staphylococcus aureus and Methicillin-resistant
Staphylococcus aureus (MRSA) and also determining their anti-topoisomerase I activity. Pure
strains were fermented in yeast extract peptone dextrose (YPD) broth for 21 days using shaker
with speed of 150 rpm at room temperature. After fermentation had finished, mycelium and
broth were separated by vacuum filtration. Mycelium was extracted by maceration and broth was
extracted by using liquid-liquid extraction. Extracts were concentrated using rotary evaporator
and then the extracts were dried in room temperature. The extracts were tested for their
antibacterial activities against Staphylococcus aureus and MRSA. Their antibacterial activities
against Staphylococcus aureus and MRSA were determined by agar diffusion method while their
minimum inhibitory concentration (MIC) were determined by broth microdilution accompanied
by minimum bactericidal concentration (MBC) determination. Anti-topoisomerase I activity of the
extracts were determined by measuring their diameter of inhibition against Saccharomyces
cerevisiae strain 1140, accompanied by calculating its IC12. Extract mycelium of GK4 showed the
biggest antibacterial activity against Staphylococcus aureus (MIC = 256 µg/mL and MBC = 2048
µg/mL) and MRSA (MIC = 1024 µg/mL and MBC = 2048 µg/mL). The result of anti-topoisomerase I
activity determination showed that extract mycelium of GK5 showed the biggest activity on
inhibiting Saccharomyces cerevisiae’s growth with IC12 = 325,40 µg/mL. Therefore isolated fungi
from Pejarakan Mangrove Forest has potential to be antibacteria and anti-topoisomerase I.
