solution. Determination of zinc sulfate in tablet is needed due to efficacy, safety, and quality of
dosage form. The purpose of this study was to develop a visible spectrophotometric method for
zinc determination. Measurement of zinc by the visible spectrophotometric is based on the
formation of colored complexes with the reagent on certain condition. A various parameter such as
reagent concentration, pH, temperature, incubation time, maximum wavelength and matrix
interferences were optimized to determine optimum condition for reaction. Method validation was
performed with the consideration of validation method parameters such as linearity and range,
limit of detection (LOD), limit of quantification (LOQ), accuracy and precision. In this study, zinc
solution was reacted with xylenol orange reagent 0.0024% w/v at pH 6, incubation allowed for 15
minutes at room temperature and the product was measured at 570 nm. Zinc that measured by
this method gave linearity in the range of 1.2 ppm - 4.8 ppm with of 0.999 coefficient of correlation,
1.3% coefficient of variance, 0.137 ppm LOD and 0.415 ppm LOQ. The accuracy value which showed
by recovery was in the range of 99.13% - 101.98%. Intraday and interday precision gave the
coefficient of variance values less than 2%. Method is used to determine the levels of zinc in
commercial tablet. The result of zinc determination in the commercial tablet fulfilled the compendia
requirements. The visible spectrophotometric method with xylenol orange had been optimized,
validated and could be used to determine the zinc level in tablet.
