This study aimed to find an optimum method of extraction and fractionation of cherry leaves based on its antioxidant activity. Cherry leaves were extracted using two methods of extraction, which were maceration and reflux. The extract that had the highest antioxidant activity was fractionated by two methods, which were liquid- liquid extraction and vacuum liquid chromatography. The optimum method was determined based on IC50 DPPH (2,2-diphenyl-1-picrylhydrazyl) reduction activity using spectrofotometer ultraviolet - visible light. The highest antioxidant activity was given by the ethanol extract with the IC50 value was 37.60 ?g/mL. Levels of total flavonoids, total phenolic and total carotenoid of cherry leaves extract were determined to analyze the correlation between these classes of compounds with their IC50 of DPPH reduction activity. The highest levels of total flavonoid, total phenolic, and total carotenoid were given by ethyl acetate macerated extract which was 20.18 g QE/100 g, n-hexane extract reflux which was 13.80 g GAE/100 g and n-hexane extract reflux which was 13.03 g BE /100 g. The statistical analysis showed no correlation between total flavonoid content, total phenol and total carotenoids with IC50 DPPH reduction activity. The highest antioxidant activity was showed by ethyl acetate fraction from liquid-liquid extraction with the IC50 value was 8.67 ?g/mL. The optimum extraction and fractionation of cherry leaves quantitatively based on its antioxidant activity were reflux and liquid-liquid extraction method. text |
