supply to the myocardium, which results in myocardial infarction. Myocardial infarction is a clinical
manifestation of coronary heart disease. Furthermore, biomarkers such as myeloperoxidase (MPO)
can give a prognosis of myocardial infarction. MPO is produced by neutrophils and macrophages
during an inflammation and is found in various stages of development of atheroma plaques. This
study aims to determine the effect of hypercholesterolemia in the inflammatory processes in the
aorta and the heart of myocardial infarction rat. First, the animals were classified into a group of
normal, normal-myocardial infarction, hypercholesterolemia, and hypercholesterolemia-myocardial
infarction. Hypercholesterolemia was induced by feeding high-cholesterol diet as well as the oral
administration of cholesterol, cholic acid and propylthiouracil. Model of myocardial infarction was

made by injecting isoproterenol intraperitoneally, one day before the animals were sacrificed. All
lipid profiles were measured in the third month, and total cholesterol were also measured before
induction, first and second month after the induction. Successful induction hypercholesterolemia,
showed by the increase of total cholesterol significantly in hypercholesterolemia group towards
normal group. MPO characterization was then performed by dot blot method. The results showed
that in the aorta of hypercholesterolemia rat, MPO expression increased significantly compared to
the normal group. The highest expression of MPO in the aorta was observed in hypercholesterolemiamyocardial infarction rat. In the heart organ, myocardial infarction animals and hypercholesterolemia
animals showed similar increase of MPO expression (1.7 times higher). However,
hypercholesterolemia-myocardial infarction rats still had the highest MPO expression (2.2 times
higher). This study suggests that high levels of cholesterol exacerbate inflammation in aorta and heart
of myocardial infarction rat which are characterized by the increase of MPO expression.
