Indonesian. Province in Indonesian which has high prevalence of T2DM is West Sumatera. Single
Nucleotide Polymorphism (SNP) rs11196205 Transcription Factor 7 Like 2 (TCF7L2) gene has been
associated with T2DM in several countries like Icelandic, Danish, United States, and Thailand. The
aim of this research was to find an association between SNP rs1196205 of TCF7L2 gene with
T2DM in ethnic Minangkabau. Design of study was a case control study included 25 T2DM
patients and 25 healthy controls who match with the inclusion and exclusion criteria.
Amplification Refractory Mutation System Polimerase Chain Reaction (ARMS PCR) was used for
identifying TCF7L2 rs11196205 gene polymorphism. Genotype of sample was determined by two
ARMS PCR reaction to detect G allele and C allele, produce 3 fragments of 856bp, 559bp, and
339pb. Data was analyzed with chi-square test. Based on the research, C allele and GC genotype
were significantly associated to T2DM (p= 0,017 for both analysis). Therefore, SNP rs11196205
TCF7L2 gene can be used as genetic marker of T2DM in ethnic Minangkabau.
