Dispersing SAP in oil is an alternative to enhance its penetration through the skin. The objective of this study is to improve dispersibility of SAP in oleid acid that can be developed further into topical nanoemulsion. Several combinations of surfactant, Tween 80-Span 80, Tween 80-Span 20, Span 80, and Span 20, were evaluated to increase dispersibility of SAP in oleic acid, that made by co-lyophilization. The formula of SAP solid-in-oleic acid dispersion that resulted in transparent dispersion were obtained with addition of Span 80 with ratio 20:1 to the SAP. Then SAP solid-in-oleic acid dispersion was incorporated into oil phase of nanoemulsion formula that stabilized by Tween 80 as surfactant and polyethylene glycol 400 (PEG 400) as cosurfactant. SAP solid-in-oleic acid dispersion was evaluated by measuring antioxidant activity and determining the dispersion type. Nanoemulsion was evaluated by measuring of globule size and distribution, determining amount of SAP in oil phase, measuring antioxidant activity using DPPH method, and in-vitro skin penetration testing by Franz diffusion cell using Python reticulatus membrane. The optimum formula of nanoemulsion containing SAP solid-in-oleic acid dispersion consisted of oleic acid, Tween 80, PEG 400, and SAP dispersion in the ratio of 9.4%, 31%, 15%, and 4.8%. The incorporation of NAP solid-in-oleic acid dispersion into nanoemulsion provided a non significant globule diameter enhancement from 175.73 ± 23.42 nm to 181.17 ± 9.13 nm (p>0.05). The scavenging activity of nanoemulsion containing SAP dispersion was 88.07 ± 1.22%. Nanoemulsion containing SAP solid-in-oleic acid dispersion enhanced its penetration through the skin compared to nanoemulsion containing SAP without dispersing treatment (p<0.05). text
