Summary:Red yeast rice is rice fermented by Monascus purpureus. Metabolites of Monascus purpureus are potential for anticholesterol and antibacterial agent. The objectives of this research was to analyze Monascus pigment that has antimicrobial activity, using bioautography method. Red yeast rice was extracted by ethanol 95% using reflux. Red yeast rice ethanol extract was fractionated by vacuum column chromatography and showed three main colours, red, orange, and yellow. The antimicrobial activity were tested by bioautography method and used Staphylococcus aureus, Escherichia coli, and Candida albicans as the tested microorganisms. Fractions which showed positive result for bioautography then refractionated by preparative thin layer chromatography. The absorbances of the subfractions were measured by UV-Vis spectrophotometry. Yellow and orange pigments gave inhibition zone against Staphylococcus aureus, but no inhibition zone against Escherichia coli and Candida albicans. Yellow and orange pigments of Monascus purpureus on red yeast rice has antibacterial activity.
