Summary:Analysis theophylline and salbutamol sulphate using spectrophotometry ultraviolet derivative has not been reported. This research aimed to apply the analysis method of spectrophotometry ultraviolet derivative in the assay mixture of two compounds that have overlapping spectrum and obtain a valid alternative method. The research was performed using the concept of derivatization with zero-crossing method. The first to second derivative was used in research and determined zero-crossing wavelength for each compound. Derivative and wavelength that used for analysis was validated. In the assay of theophylline and salbutamol sulphate with a combination of 130 : 1 using zero-crossing method, only theophylline assay that could be determined with a valid result. Analysis of theophylline performed on first derivative with a wavelength at 245,2 nm. The average recovery study of theophylline was 99,1%, 100,11% and 99,85% with relative standard deviation values for intraday precision was 0,83%, 1,43% and 1,62%. For interday precision values for each concentration was 1,3%, 1,17% and 1,4%. From this research the conclusion is the analysis method of spectrophotometry ultraviolet derivative only could applied for determination of theophylline in combination tablet with providing good accuracy and precision.
