This research aimed to apply the analysis method of spectrophotometry ultraviolet derivative in the assay mixture of two compounds that have overlapping spectrum and obtain a valid alternative method. The research was performed using the concept of derivatization with zero-crossing method. The first to second derivative was used in research and determined zero-crossing wavelength for each compound. Derivative and wavelength that used for analysis was validated. In the assay of theophylline and salbutamol sulphate with a combination of 130 : 1 using zero-crossing method, only theophylline assay that could be determined with a valid result. Analysis of theophylline performed on first derivative with a wavelength at 245,2 nm. The average recovery study of theophylline was 99,1%, 100,11% and 99,85% with relative standard deviation values for intraday precision was 0,83%, 1,43% and 1,62%. For interday precision values for each concentration was 1,3%, 1,17% and 1,4%. From this research the conclusion is the analysis method of spectrophotometry ultraviolet derivative only could applied for determination of theophylline in combination tablet with providing good accuracy and precision. text
