Summary: | Acyclovir is a synthetic purine nucleoside analogue for the treatment and prophylaxis of
infections due to herpes simplex virus types 1 and 2 and varicela-zoster virus. Some
quantitative analytical methods of acyclovir have been reported using high performance
liquid chromatography and UV-Vis spectrophotometry. However, this methods need many
toxic solvents, more consuming time and relatively expensive. The aim of this research is to
develop FTIR as more rapid and cheaper alternative method for determination acyclovir in
acyclovir tablet. Infrared spectrum of acyclovir was changed to be a first derivative spectra,
then measured the AUC (area under the curve). Calibration curve was made in concentration
of 0.2-1.2% w/w and wavenumber was optimized at 3700-3440, 3012-2870, 1955-1680,
1176-1072, and 721-609 cm
-1
. Analytical method in this research was validated by
specificity, linearity, limit of detection, limit of quantification, accuracy and precision test.
After this method had good validated, amount of acyclovir in tablet were measured. The
optimal wavenumber in this research was 3700-3440 cm
-1
and produced a regression equation
y = 54.922x + 17.479 with correlation coefficient (r) 0.9999 and variation coefficient of linear
regression (Vxo) 0.45%. Specificity test showed that this method was specific. The limit of
detection and limit of quantification was obtained at 0.01 and 0.03 % w/w. Recovery of
accuracy test was 99.98-101.21%. Relative standard deviation of precision test was 0.27-
0.38% and Horrat value was 0.07-0.09. Acyclovir tablet of sample A, B, and C contained
acyclovir respectively 103.49-103.77; 101.27-102.33; and 98.92-100.02%. All of the result of
this research showed that FTIR can used as good validated alternative method, more rapid
and cheaper for determination acyclovir in acyclovir tablet.
