The release of cytokines, especially TNF-?, caused synovial inflammation leading to destruction of cartilage and joints which are the characteristics of RA. Analgesic and anti-inflammatory drugs are used to treat RA but long term usage of such medication is associated with adverse reactions and discomfort. Natural plant products that are beneficial against arthritis are continously being sought for the management of RA. The objective of this research is to examine the anti-rheumatoid arthritis activity of water extract of green tea [Camellia sinensis (L.) Kuntze] and avocado (Persea americana Miller) leaves which is stated in literature have anti-inflammatory activity. The research was begun with collecting, determining and characterization of the test plant, extracting the active substances in water as traditional method, modelling of CIA rats, followed by testing those extracts against CIA rats modelled. The presence of antiarthritic effect was determined through arthritis or macroscopic scoring, TNF-? concentration, and histopathological study of rat’s limb tissue. Through arthritis scoring, after the CIA rats treated with the two doses of green tea (72 and 216 mg/kg) and avocado leaves (44 and 88 mg/kg) extracts, there was a decreased in macroscopic scoring. Under TNF-?concentration determination, green tea at dose 216 mg/kg and the two doses of avocado extracts reduced TNF-?concentration significantly (p<0.05). Through microscopic examination of hind limb rat joint tissue, green tea at dose 216 mg/kg could supress cartilage erosion better than other. Based on the results, it can be concluded that water extract of green tea at dose 216 mg/kg has an anti-rheumatoid arthritis in CIA rat model. text |
