that included in Tephrosia are quersetin, flavanones, isoflavones, flavonols, and rotenoids.
The aim of this research was to isolate flavonoid compound from Tephrosia vogelii leaves.
Crude drug was extracted by reflux using methanol as solvent. Methanol extract was
fractionated by liquid –liquid extraction method (charaux-paris method) and using radial
chromatography. Purification was done by dissolving targeted compound using suitable
solvent. Isolate was characterized by spectrophotometer UV –visible with shifting reagent
and spectrophotometer IR. Phytochemical screening indicated that crude drug of Tephrosia
vogelii leaves contained alkaloid, flavonoid, and steroid/triterpenoid. Methanol extract
contained alkaloid and flavonoid. Isolated compound gave positive result with citroboric
and AlCl3 reagent. The UV spectrum of methanolic solution of isolate exhibited two major
absorption bands at 312 nm and 270 nm. After addition of shifting reagent the wave length
shifted to 317 nm and 275 nm when it was added with sodium acetate. In other hand, the
result of spectrophotometer IR had hydroxyl group at 3000 –3300 cm
-1
, C=O at 1700 cm
-1
,
and aromatic group at 1500 cm
-1
. Isolated A was proposed as isoflavone with hydroxyl
group at C-7.
