Curcumin has been widely used as an active ingredient that has the potential as an antioxidant, anticancer, anti-inflammatory, antimicrobial, antifungal and antiviral. The main constraints in the development of curcumin preparations are their low solubility in water, degraded by light and temperature, easily metabolized to inactive forms that causing low bioavailability of the body, and high metabolism in the liver. Curcumin has been tested to determine the effect of dengue antivirus, specifically on the DENV-2. The dengue virus has a genome that encodes three structural proteins and seven nonstructural. This study aims to develop nanoemulsion containing curcumin (curcumin nanoemulsion) and evaluate physical characteristics, cytotoxicity effects of curcumin on A549 cells and selectivity of inhibitory effects on dengue virus in serotypes 1 and 2. The cell cytotoxicity effect of curcumin nanoemulsion was determined using the MTT assay on the A549 human lung cancer cell line to obtain a CC50 value, compared with a curcumin solution in DMSO. Inhibitory effect on DENV1 and DENV-2 virus titers with the RT-qPCR method to obtain an IC50 value. Curcumin nanoemulsion was successfully formed by emulsification process, resulting in nanoemulsion with a uniform droplet size distribution. The results of the droplet size characterization of curcumin nanoemulsion were 24,8 ± 2.5 nm, the polydispersity index of curcumin nanoemulsion was 0.189 ± 0.385 and the potential zeta of curcumin nanoemulsion was -4.3 ± 0.5 mV. The morphology of encapsulated curcumin in the nanoemulsion system has a spherical and uniform shape. The average percentage of the loading capacity of curcumin was 93.750 ± 0.950%. The CC50 value of curcumin nanoemulsion obtained was 21,09 ± 1,38 µg/ml, while the CC50 value of curcumin solution obtained was 33,66 ± 5,91 µg/ml. IC50 value of curcumin nanoemulsion was 1.688 µg/ml for DENV-1 and 4.809 µg ml for DENV-2. Curcumin in the form of nanoemulsion and solution in DMSO is better in reducing DENV-1 virus titer compared to DENV-2. However, curcumin in the form of nanoemulsion and solution is very responsive in decreasing the virus titer in DENV-1 and DENV-2. text
