If this concentration is too high, uric acid will form a crystal. This crystal will be deposited in joints and when this happened, it is called gout. First line therapy for this disease is allopurinol. Allopurinol is an inhibitor of the enzyme xanthine oxidase (XO). However, this medicine has Steven-Johnson Syndrome as a side effect from consuming it. Therefore, alternative medicine that has similar activities needs to be searched to reduce the side effects. Cherry leaves has the XO inhibitor. The objective of this experiment is to isolate the substance that has XO inhibitor activity. Extraction of cherry leaves from three different areas, that is from Bogor, Sumedang, and Magetan, used maceration method with ethanol 96%. Fractionation was done by liquidliquid extraction with n-hexane and ethyl acetate. Subfractionation was done by vacuum liquid chromatography with eluent that increasing polarity. Purification was done by preparative thin layer chromatography. Ethanol extract of cherry leaves from Bogor can inhibit 23,46% of XO in 100 µg/mL. Ethyl acetate fraction can inhibit 34,66% of XO in µg/mL. The purification results with preparative thin layer chromatography, four isolates are obtained, one of the isolates is pure. text
