Phytochemical characteristic and bioactivities of honey depend on their collected
location, flower sources, bee species and industrial processes. The purpose of this
study was to determine antioxidant activities of Apis cerana honey from Badung,
Karangasem and Singaraja Districts in Bali Province, Indonesia by using DPPH and
FRAP methods. Also, total phenolic and flavonoid content of honey samples were
determined, and correlations between these parameters were evaluated. Methods:
Honey was extracted by reflux technique consecutively using n-hexane, ethyl
acetate then ethanol and directly using ethanol. Antioxidant activities, total phenolic
and flavonoid were identified with UV-Vis spectrophotometer. In addition,
antioxidant activitiy index (AAI) of honey extracts were determined. Correlation of
total phenolic and flavonoid content with AAI of DPPH and FRAP also the
relationship between two antioxidant methods were analyzed by Pearson’s method.
Result: The highest antioxidant activities by DPPH and FRAP were shown by ethyl
acetate extract of Karangasem honey with AAI values were 33.521 and 4.874
respectively. This sample also showed the highest content of phenolic (4.327 g
GAE/100 g) and flavonoid (2.053 g QE/100 g). Total phenolic content of all
samples had positively and significant correlation with their AAI of DPPH and
FRAP. AAI of DPPH from Karangasem and Singaraja honey gave positively and
significant correlation with their AAI of FRAP. Conclusions: All extracts had
antioxidant activities. Phenolic compounds mainly contributed in antioxidant
activities of honey extracts by DPPH and FRAP methods. DPPH and FRAP
methods showed liniear results in antioxidant activities of honey extracts from
Karangasem and Singaraja.
