The method need three component for diagnosing, the component are blood sample / serum from the patient, diagnostic kit and ELISA reader instrument. This research designed an ELISA reader that is composed of an LED connected to an optical fiber as its light source, silicone photodiode as its sensor, stepper motor as wellplate driver and microcontroller as controller. This research uses one LED lamp with a wavelength of 450 nm. A mechanical construction is designed to connect LED lamps with optical fiber. The purpose mechanical construction are to split one light source into eight light sources, so that each sensor receives the same wavelength. A total of eight SFH213 photodiode sensors were used for measurements. The measurement validation was carried out by using the benchmarking method against commercial ELISA readers. The commercial ELISA reader used is iMark from Biorad. The test results prove that the ELISA reader being designed could measure from 0 OD to 1,6 OD with an error of 0.093 ± 0.098 OD. Based on these results, the ELISA reader meets the specifications to be used to detect hepatitis B virus infection. text
