Summary: | The combination of two active pharmaceutical ingredients in a single
pharmaceutical dosage form can cause intermolecular interactions. Intermolecular
interactions may affect the physicochemical properties of the constituent
components. Identification of intermolecular interactions of the mixture of
telmisartan (TMS) and amlodipine besylate (ADB) was carried out by the fusion,
solid state grinding (SSG), solvent drop grinding (SDG) and solvent evaporation
(SE) method in the 1:1 ratio molar. The fusion method in methanol solvent showed
a new crystalline habit indicating the presence of intermolecular interactions. The
TMS-ADB phase diagram indicated the occurrence of peritectic interactions. The
DSC thermogram showed the presence of two endothermic peaks which are the
melting points of the peritectic compound. Microscopic analysis by SEM of SDG
method showed the agglomeration in the TMS-ADB mixture. The IR spectrum
showed a shift in the wave number in the O-H and C = O stretch. X-ray
diffractograms did not show any new crystalline phase. The solubility of the TMS
after treatment in phosphate buffer medium pH 7.5 increased up to 3 times
compared to its pure form.
