This study aims to determine the blood pressure-lowering activity of ethanolic extract of papaya (Carica papaya L.) leaves in Sprague Dawley male rats to test the diuretic and antihypertensive activity with adrenalineand NaCl inductors. Diuretic activity tested with metabolic cages performed in the control group, furosemide 1,8 mg/kg bw, and extract doses of 125 mg/kg bw, 250 mg/kg bw, and 500 mg/kg bw. The entire group was given a 50 mL/kg physiological saline as loading dose. Except for control group, it also given a dosage of furosemide or test extract. Parameter of percent volumetric urinary excretion was observed every hour for 6 hours and accumulated up to 24 hours. Antihypertensive activity tested with adrenaline and NaCl inductors made on the negative control, positive control, and extract with three different doses and comparative, respectively, atenolol 4,5 mg/kg bw and captopril 13,5 mg/kg bw. Parameters such as amplitude and heart rate (frequency) at adrenaline induction observed in T-0 before the given dosage, T-1 after a given dosage, as well as T-2 after adrenalin induction intraperitoneally, while the parameters of NaCl induction observed at day 0 before induction, day 14 after NaCl induction orally, and day 28 after induction and therapy. Diuretic test showed that ethanolic extract of papaya leaves have diuretic effect on doses of 125 mg/kg bw, 250 mg/kg bw, and 500 mg/kg bw in compare to furosemide 1,8 mg/kg bw on the 2nd hour of observation. Antihypertensive test with adrenaline induction showed the effect of decreasing the amplitude of the heart rate of ethanolic extract of papaya leaves doses of 125, 250, and 500 mg/kg bw comparable with atenolol dose of 4,5 mg/kg bw. Extract 250 mg/kg bw has the same effect with atenolol dose of 4,5 mg/kg bw in inhibiting the increase of heart rate induced by adrenaline. Antihypertensive test with NaCl induction showed the effect of decreasing the amplitude of the heart rate of ethanolic extract of papaya leaves doses of 125, 250, and 500 mg/kg bw comparable with captopril dose of 13,5 mg/kg bw. NaCl induction could not increase the heart rate so that the effect of decreasing the frequency of the captopril and the exctract could not be observed. text |
