Summary: | Katuk (S. androgynus) has been used traditionally in India as diabetic therapy, known
as “green diabetic”, but the compound that was responsible for that effect was
unknown. The aim of this research were to isolate and to elucidate the structure of
chemical compound which inhibit ?-glucosidase enzyme. Katuk Leaves were extracted
by maceration using ethanol 70%. The extract were fractionated by liquid-liquid
extraction using n-hexane, EtOAc obtained three different fractions. All of the extract
and fractions were screened for ?-glucosidase inhibitory activity. The highest activity
were found in EtOAc’s fraction. It was continued for further separation using vacuum
liquid chromatography. FE-SA-01 (110 mg) was successfully isolated using radial
chromatography. The structure determination has been done based on spectroscopic
method (UV, IR and NMR/RMI). Based on the characterization’s data the isolated
compound was proposed as Astragalin. The IC50 was 55.3 ± 0.61µg/mL compared to
acarbose as positive control which was 162.96 ± 0.59 µg/mL. This is the first report for
isolating Astragalin in Katuk Leaves.
