Summary: | Groundnut shell is an abundant waste produced in Indonesia with minimum utilization. The high cellulose content of groundnut shells makes it a suitable substrate for cellulase production. Cellulase production has been considered as an expensive process because of the high cost of pure cellulose that is used as substrate for fermentation. The use of groundnut shells as abundant and cheap raw material for cellulase production can be an option for solving aforesaid problem. This study aimed to determine the effect of solid-sate fermentation time using Trichoderma sp. on cellulase activity and determine optimal fermentation time. The fermentation process was carried out at room temperature (25?), 50% humidity inside the fermentation flask with a substrate to culture ratio of 10:1 for 5 days with measurement of enzyme activity every 24 hours. FPase method was chosen to determine the enzyme activity. The values of cellulase activity for 1, 2, 3, 4 and 5 days of fermentation were 0.02; 0.05; 0.07; 0.08; and 0.02 FPU/ml, respectively. In addition, mathematical modeling was also carried out using secondary data to determine the kinetic parameters of cellulase production process for SMF and SSF fermentation method. The value of kinetic parameters ?m, Ks, kd, ms, Yx/s, k1, k2, and Ki for SMF method were 0,22 h-1; 29,73 g/l; 0,0736 h-1; 0,0 006 h-1; 1,19 g/g; 0,9429 IU/g h; 0,1062 h-1 and 1138 g/l respectively. Meanwhile the value of kinetic parameters ?m, Ks, kd, ms, Yx/s, k1, k2, and Ki for SSF method were 1,9 h-1; 6,36 g/l; 0,1691 h-1; 0,0001 h-1; 26,01 mg/g; 0,1896 IU/ g h; 0,0516 h-1 and 0,0013 g/g respectively. |
