Therefore, an effort is needed to control dengue virus infection. The dengue virus consists of four serotypes, namely DENV- 1, DENV-2, DENV-3,and DENV-4. DENV-3 is the most infecting dengue serotype in Bandung. Envelope proteinis one of the important proteins in the dengue virus. This protein functions to bind to the receptor region and fuse the virus with the host cell membrane. In addition, envelope proteinalso plays a role in stimulating the immune system through the formation of neutralizing antibodies. This ability makes the envelope protein can be used as an antigen in the development of antibody-based dengue infection detection kits. The purpose of this study was to model the envelope protein structure of dengue virus serotype 3 isolated from Bandung and analyze the epitope of the protein. Then after the epitope was identified, the envelope protein of DENV-3 was expressed in Pichia pastoris cells. Protein structure modelling was carried out using SWISS MODEL servers with a residue gained in the most preferred region is 84,6%. Linear epitope analysis was performedusing the Bepipred Linear Epitope Prediction 2.0, Chou and Fasman Beta-Turn Prediction,Kurplus and Schulz Flexibility Prediction methods, and also using BCPred. Meanwhile, the discontinuous epitope was analyzed using two methods, namely Discotope and Epipred. Linear and discontinuous epitope on the envelope protein DENV-3 Bandung Isolate were successfully determined andare estimated to have several sites scattered throughout the domain. The results of DENV-3envelope protein expression in Pichia pastoris cells observed using the SDS PAGE techniqueshowed a protein band with size 45 kDa. text
