Summary: | Ascorbic acid is one of the essential water-soluble vitamins needed by the body and acts as an antioxidant, collagen former, and maintains body immune systems. Ascorbic acid is not stabilized by the presence of light, oxygen, and metals. This study aims to develop validated analytical methods to determine the levels of ascorbic acid in commercial healthy beverage products and to compare the results of testing the suitability of ascorbic acid levels in samples with levels indicated on the label. Detector used is UV-Vis at 244 nm and Sunfire C-18, 5gm, 4.6 x 150 mm column. High performance liquid chromatography system was optimized by testing the suitability of the system and further validated. Validation was conducted on the following parameters: specificity, linearity, limit of detection, limit of quantification, accuracy, and precision. Optimization method was obtained from mobile phase of acetonitrile: 1% metaphosphoric acid (30:70) pH 2.5, flow rate of 0.6 mL/min, column temperature of 25°C. Value of the correlation coefficient (r) obtained was 0.999; linear regression coefficient of variance (V„,„) of 0.17%; limit of detection 2.50 i.tg/mL; and limit of quantification 7.58 1.1g/mL. The first day of the test results showed precision of 0.96% coefficient of variance; the second day test at 1.12% and 0.70% for the third day. Accuracy of test results for samples 1, II and III, respectively, showed the recovery of 100.87%, 99.37% and 97.03%. The method of analysis using high- performance liquid chromatography used could be done to analyze the level of ascorbic acid in various samples of beverages. Levels in samples A, B, C, D and E were 103,57%, 134,51% 88,03%, 95,51%, 116,43%, respectively. |
