To produce biomass, the microalgae were cultivated through batch culture with aeration using air or CO2. Prior to lipid extraction, microalgae cells were pretreated using ultrasonic method or freeze-drying. The lipid content was extracted in soxhlet apparatus using organic solvents of hexane or chloroform:methanol (1:2). The fatty acid contents were characterized by the GC-MS analysis. The biomass concentration obtained from cultivating with aeration using air and CO2 was 2.63 and 0.32 g/L, respectively. The total of fatty acid content obtained from cell cultivated with aeration using air and CO2; from cell preteated by sonication and freeze-drying; and from lipid extraction using hexane and chloroform:methanol (1:2) was 65.5 and 53.9%; 65.5 and 30.5%; and 29.9 and 30,5%, respectively. The results indicate that the optimum condition for fatty acids production occur when cells are cultivated with aeration using air and pretreated by sonication prior to extract their fatty acid using chloroform:methanol (1:2) solvent. text |
