Summary:The purpose of this research is to isolate, purify and characterize sulfhydryl protease (SH-protease) from Pseudomonas cocovenenans X-128, as the first step to reveal the enzymology of bongkrekic bacterium. Most of the work which have been done so far deal with the two toxic substances toxoflavin and bongkrekic acid produced by this bacterium. Where as to our knowledge no work on the enzymology of this bacterium which often causes poisoning has been carried out except for this research. In the preliminary study we found that the bongkrekic bacterium produced intracellular SH-protease. The existence of this enzyme in this bacterium has attracted attention because in vitro bongkrekic acid inhibits the SH-protease while in vivo does not kill bongkrekic bacterium. Generally SH-proteases are only found in certain plants and are rarely found in micro-organism. Protease is also known to play an important role in metabolism, scientific and indastry. For this reason SH-protease from Pseudomonas cocovenenans X-128 was isolated, purified and characterized.
