This study aims to determine the concentration of 2,4-dichlorophenoxyacetic acid (2,4-D) and benzylaminopurine (BAP) to induce callus culture and cell suspension of A. malaccensis. Callus initiated from 1 year old young shoots of A. malaccensis on solid Murashige Skoog (MS) medium with the addition of 0.3; 0.4; and 0.5 ppm 2,4-D and BAP with a concentration of 1, 1,5; and 2.0 ppm. The results showed that a combination of 0.5 ppm 2,4-D and 2.0 ppm BAP and a combination of 0.4 ppm 2,4-D and 1.5 ppm BAP produced the best response in inducing callus formation. Then the callus was subcultured into liquid MS medium to induce the formation of a cell suspension culture. Quantitative parameters in the form of cell concentrations were observed weekly for a period of 4 weeks to observe the growth curve, doubling time, and specific growth rate. The combination of 0.4 ppm 2,4-D and 1.5 ppm BAP was able to induce the formation of a cell suspension culture with a specific growth rate of 0.062 and a doubling time of 11 days, while the combination of 0.5 ppm 2,4-D and 2.0 ppm BAP was able to induce the formation of a cell suspension culture with a specific growth rate of 0.031 and a doubling time of 22 days. Observation of cell morphology showed that the suspension culture on a combination of 0.4 ppm 2,4-D and 1.5 ppm BAP was dominated by single cells with a percentage of rounded cells of 91.67%; and the percentage of oval cells was 8.33%, whereas in the combination of 0.5 ppm 2,4-D and 2.0 ppm BAP, suspended cells were dominated by single cells with a percentage of round cells 89.65%; and the percentage of oval cells is 10.35%. The combination of 0.5 ppm 2,4-D and 2.0 ppm BAP stimulated more cell growth in the 4th week but with a lower percentage of rounded cells than the 0.4 ppm 2,4-D combination and 1.5 ppm BAP. It can be concluded that the combination of 0.5 ppm 2,4-D and 1.5 ppm BAP can induce faster multiplication of A. malaccensis cell suspension cultures than the combination of hormones 0.5 ppm 2,4-D and 2.0 ppm BAP.
