Summary: | The production of tablets is a very intricate process that is influenced by numerous
process variables or parameters. The objective of this work was to identify, using a
statistical experimental design, the critical processing variables that affect Critical
Quality Attributes (CQAs) Vitamin C film-coated caplets. A two-level complete
factorial design with two central points was used to examine the process
parameters that posed the greatest risk to CQA. The process variables investigated
were mesh number, lubrication time, compression process speed and main
thickness. Statistical results showed that mesh number, lubrication duration, and
their interaction have significant effects on flow rate, Hausner Ratio, and
compressibility index. Higher mesh number and longer duration resulted in better
flow properties. Main thickness parameter has significant effect on core caplet
hardness, thinner main thickness will increase caplet hardness. The compression
speed parameter has a significant effect on product dissolution, low product
dissolution observed due to higher speed compression. Based on this study, it can
be concluded that mesh number and lubrication duration only have a significant
impact on the bulk quality attributes, but do not have a significant impact on the
Vitamin C caplets quality attributes, while the compression process parameters
(speed and main thickness) have a significant effect on Vitamin C caplets quality
attributes. Mesh number 20 with a lubrication time of 7 minutes, 17 rpm speed with
a major thickness scale of 2.00 are estimated to be the best parameter processes
for producing optimum CQA Vitamin C caplet.
