INSTITUT TEKNOLOGI BANDUNG https://digilib.itb.ac.id/gdl/view/78315 Surface Plasmon Resonance (SPR)-based sensing has gained increasing popularity in various biochemical applications, especially for detecting biomolecular interactions due to its capability for real-time detection and proven high sensitivity for label-free detection of biomolecules. Consequently, there is a growing demand for the development of SPR sensor technology for rapid testing at point-of-care. However, commercialized SPR sensors are not unfitting to enable rapid testing at point of care due to their large size, lack of portability, and high cost. On the other hand, fiber optic-based SPR sensing offers the potential for compact and portable SPR devices, well-suited for easy, fast, and cost-effective point-of-care testing. In this final project research, the author has conducted an experiment as a preliminary study to develop an SPR sensor using polymer optical fiber (POF) and optimization of fabrication techniques and thin gold film thickness deposition was conducted to characterize the optical parameters of the POF-based SPR sensor in response to changes in the refractive index using varying concentrations of glycerin solutions. From a range of POF-based SPR sensors variation, the best-performing sensor achieving a sensitivity value of 4,585 ± 0,489 within R-squared coefficient 0,936, a Limit of Detection (LoD) of 0.31 mM, stability of 63,5%, and a penetration depth of 77.7 nm. These results demonstrate that the fabricated POF-based SPR sensor can generate surface plasmon resonance and deliver a robust SPR response in detecting changes in glycerin concentration within the sensing element with a D- shaped geometry. This research paves the way for the development of compact and portable SPR sensors suitable for point-of-care applications. text
