Summary:Background and Purpose : The coprocessed is a technique to improve the function of excipients such as compactibüity and flowability. The calcium carbonate is used as an alkaline diluent that can increase solubility and dissolution of drugs and also uses in formulas of tablets effervescent, chawable and calcitm supplement> but it's compactibility is poor. The mannitol is plastis and reported can improved the hardness of tablet and compactibility„ The aim of this research was to increase the functionality of calcium carbonate by mixing with mannitol made by spray drying and wet granulation. Methods The coprocessed mannitol-calcium carbonate was made with drying (SD-MC) and wet granulation (GB-MC) at ratio of 8020; 6040; 40:60; and 20180. The determination of parameters of spray drying process used a mannitot solution of 4%, 8% and 12%, the inlet temperature 900C or 1 ITC, feed rate, air pressure pump gauge, blower flow meter are 6 grams/miny 1 kg/cm2 and 0.45 Nm3/miny respectively. The process parameters that produced the highest yield of powder was then used in the manufacture of coprocessed mannitol-calcium carbonate. The powders and granules obtained were then evaluated including the value of Carrvs index, Hausner ratio, flowability, particle size distribution, compactl%llity, X-ray diffraction, thermal analysis (TGADSC), FTR and SEM. Results : The process producing the highest yield was given by mannitot solution of 4% and an inlet temperature of HO oc The results of flowability test of coprocessed SD-MC and GB-MC were 0.90 to 1.48 g/sec and 3.82 to 7.16 g/sec, respectively. The results of tensile strength at compression force of 9.81 kN for SD-MC powder (with a ratio of80:20; 60:40•, 40:60 and 20.•80 ) were 1.37 MPa; 1.93 wa•, 0.98 MPa•, and 1.51 tvfPa, respectively- Meanwhile, the value of the tensile strength GB-MC were 0.88 MPa; 0.78 Iv1Pa; 0.61 and 0.54 Tv1Pa, sespectively. The tensile strength of coprocessed by spray drying was greater than the wet granulation at the ratio 60:40. The result of X-ray diffraction of coprocessed powder gave similar peaks with its components (mannitol and calcium carbonate) while FTIR analisys gave no interaction and the morphology by SEM its was observed that the spray drying produced the spherical particles. As addition the results of thermal analysis, showed that the enthalpy of coprocessed powder Were 169.30 J/g, 454.38 J/g and lower than their constituent. Conclusions The coprocessed mannitol-calcium carbonate by spray drying at ratio of MC 60-.40 gave a better compaction while by wet granulation gave a better now.
