Therefore, it is necessary to find an alternative treatment i.e by using herbs. This research aimed to study the activity of methanol extract ofjackfruit leaves (EMDN) and ethyl acetate fraction of breadfruit leaves (FEADS) in treating rheumatoid arthritis. The active compounds of jackfruit and breadfruit leaves were extracted by maceration using methanol. Ethyl acetate fraction of breadfruit leaves is obtained by fractionation of methanol extract using liquid-liquid extraction method. This study begins with an acute anti-inflammatory test followed by anti-rheumatoid arthritis in rat model. In the acute anti-inflammatory test, Wistar rats were induced having acute inflammation by injecting intradermally of carrageenan lambda into hind limb footpad. Animal model of rheumatoid arthritis (AR) were perfomed in Wistar rat by injecting collagen type Il bovine emulsified in CFA for immunization phase and in the IFA as booster phase. Two doses of extracts were tested i.e., 150 and 200 mg/kg body weight for EMDN, and 157.5 and 215 mg/kg body weight for FEADS. The parameters of the effect ofEMDN and FEADS in rat RA model observed i.e, arthritis score, TNF-a level and histology of joint. In the acute anti-inflammatory test, both doses of EMDN can inhibit the formation of inflammation significantly (p<0.05) i.e., 24% for dose 75 mg/kg body weight and 55.4 % for dose 150 mg/kg body weight. In the AR model test, both doses of EMDN and FEADS at a dose of 157.5 mg/kg body weight can reduce arthritis scores significantly (p<0.05) with the higest percentage of reduction i.e 41.02% occurred after administered of EMDN at dose of 150 mg/kg body weight. EMDN at dose of 150 mg/kg body weight can reduce level of TNF-u significantly (p<0.05) compared than control. Through histological examination of hind limb joints of RA rats showed an improvement Of the joint cartilage indicated by the absence of cartilage erosion after treated with the two doses of EMDN and FEADS 157.5 mg/kg body weight. Based on those results, it can be concluded that EMDN at dose of 150 mg/kg body weight have a -better activity in treating rheumatoid arthritis compare to EMDN 200 mg/kg body weight and both doses of FEADS. text |
