Jamblang is a native plant of Indonesia which include in Myrtaceae family and known as traditional medicine for curing diabetes mellitus, diarrhea, cough and inflammation. Ethyl acetate fraction of jamblang was reported to have high antioxidant activity. Catechin and ferulic acid were compounds that had a role in antioxidant activity. Other phenolic compounds such as flavonoids were known to have antioxidant activity. The purposes of this study were to isolate and identifify antioxidant compounds besides catechin and ferulic acid from leaves of jamblang. Extraction was perfonned by soxhlet using three solvents with the increasing polarity (n-hexane, ethyl acetate, and methanol). Isolation process of jamblang leave extract used vacuum liquid chromatography (VLC) and size exclusion chromatography (SEC). The antioxidant activity was measured by DPPH method coupled with ELISA microplate. IC50 of ethyl acetate jamblang leave extract, SEC subsubfraction and ascorbic acid were respectively 41,39; 8,56 and 6,65 pg/mL. Based on the monitoring by TLC with citroboric, it was known that the subsubfraction was a flavonoid. Based on the results of spectrum analysis, compound X in the methanol showed of band I at 352 nm and band Il at 257 nm. After it was given shifting reagent NaOMe, AIC13, ÄIC13/HCL, NaOAc, H3B03, the band I moved to batocromic shift respectively 363, 430; 399, 381 and 371 nm. Analysis result shown that compound X had the hydroxyl molecule (OH) at 3',4',7,5 and 3-0H substituted. Compound X was identified as quercitrin after comparison of UV specfrum and TLC data with the authentic marker. Compound X is one of antioxidant compound in SEC subsubfraction and identified as quercitrin. text |
