This aims of this research is to carry out isolation and characterized of heterotrophic microalgae as source of docosahexaenoic acid (DHA) for Mangrove Areas at Cilacap. The heterotrophic microalgae were isolated from mangrove fallen leaves (Rhizophora apiculata) using direct planting method, The morfology of microalgae colony were observed through light microscope and then it was purified to produce single colonies that is subsequently fermented for 14 day to produce biomass. Fatty acids were extracted and methylated through direct transesterification method. The content of DHA by was using determination Gas Chromatography. The result were four isolates heterotrophic microalgae MTKCI, MTKC2, MTKC3 and MTKC4. The isolate of MTKC2 which only detected produced DHA with value of The MTKC2 was subsequently were identified and characterized by molecular biology. The identification results showed that MTKC2 include genus Thraustochytrium. text
