Summary:Etoricoxib is one of the COX-2 selective anti-inflammatory groups classified in BSC class II. In this study, an attempt was made to increase the solubility of etoricoxib through a solid dispersion method using Kollidon® VA 64. This experiment began with a preliminary test to find the optimal comparison between etoricoxib and Kollidon® VA 64. After the test, it was found that the ETO-KOL-3 (1:3) ratio was optimal. Then, a solid dispersion was made using a solvent evaporation method which was divided into three, namely drying the solvent at room temperature (ETO- KOL-3-SR), drying the solvent with a heating rate (ETO-KOL-3-SE) and drying the solvent with Spray Drying (ETO-KOL-3-SD). The results of the solid dispersion were then characterized by Powder X-Ray Diffractometry (PXRD), Differential Scanning Calorimetry (DSC) and Scanning Electron Microscope (SEM). The PXRD results of ETO-KOL-3-SR, ETO-KOL-3-SE and ETO-KOL-3-SD have a degree of crystallinity of 37.62, 36.72 and 35.55. The DSC results of ETO-KOL-3- SR, ETO-KOL-3-SE and ETO-KOL-3-SD have a Tg of 72.78, 80.87 and 88.48°C. The SEM results of ETO-KOL-3-SR and ETO-KOL-3-SE have an irregular plate shape but ETO-KOL-3-SD has a spherical shape. The dissolution results of ETO- KOL-3-SR, ETO-KOL-3-SE and ETO-KOL-3-SD in phosphate buffer media pH 6.8 have a dissolution percentage of 53.39±0.34%, 63.05±0.24% and 72.60±0.19%. In this study, it can be proven that the formation of solid dispersion of etoricoxib with Kollidon® VA 64 by Spray Drying method is one of the potential approaches to increase the dissolution rate of etoricoxib.
