Summary:Currently, the demand for large-scale production of patchouli plants has increased. This is because patchouli plants have become the main source for obtaining patchouli essential oil, which is widely needed in various industries such as aromatherapy, fragrance production, cosmetic products, soap and shampoo manufacturing, as well as a preservative in the food industry. However, conventional patchouli cultivation in open areas is highly susceptible to pests and diseases, and there are inconsistencies in oil quality. An alternative in vitro seedling cultivation can be done in mist bioreactors to increase patchouli plant production and essential oil biomass. Product enhancement can also be achieved through elicitation techniques. Patchouli cultivation in mist bioreactors is carried out for 14 days with a watering system of 15 minutes every 1 hour and the addition of methyl jasmonate (MeJA) elicitor at 10 µM for 72 hours. This research aims to enhance seedling growth and essential oil content of patchouli plants through tissue culture by applying methyl jasmonate elicitor treatment to plantlets cultivated in mist bioreactors. Results show that the biomass of control patchouli and 10 µM MeJA treatment increased by 47.31% and 60.5%, respectively. The growth rate & RGR of control patchouli and 10 µM MeJA treatment were 0.143 cm/day; 0.073 g/day and 0.145 cm/day; 0.086 g/day, respectively. The survivability rate for both variations was 100%. Patchouli oil extraction was performed through steam distillation for 6 hours. Oil yields for control patchouli and 10 µM MeJA treatment were 8.078% and 11.52%, respectively. Thus, it can be concluded that the use of 10 µM MeJA elicitation can enhance seedling growth and essential oil content of patchouli plants
